Synthesis of UDP, UTP, and CTP

For medical students2 min readUpdated 2026-10-10

The biosynthesis of cytidine triphosphate (CTP) and uridine polyphosphates is a sequential chain of reactions converting monophosphates into active triphosphates. ATP plays a crucial role in these processes as a universal energy and phosphate group donor, alongside specific kinases and synthetases that modify the pyrimidine ring.

Formation of UDP and UTP

The synthesis of uridine polyphosphates represents the sequential phosphorylation of monophosphates. This process proceeds in two stages, each catalyzed by a specific enzyme from the kinase class. The universal phosphate group donor in both reactions is an ATP (adenosine triphosphate) molecule. During the reaction, the $\gamma$-phosphate residue of ATP is transferred to the corresponding nucleotide.

  1. The first stage is the conversion of uridine monophosphate (UMP) into uridine diphosphate (UDP). The reaction is catalyzed by the enzyme NMP kinase (nucleoside monophosphate kinase). The reaction equation is as follows:

UMP + ATP → UDP + ADP

  1. The second stage consists of the further phosphorylation of UDP to uridine triphosphate (UTP). The enzyme NDP kinase (nucleoside diphosphate kinase) is responsible for this step. The reaction proceeds via a similar mechanism utilizing ATP energy and phosphate:

UDP + ATP → UTP + ADP

Synthesis of CTP (Cytidine Triphosphate)

Once UTP has formed within the cell, it can be used as a substrate for the biosynthesis of cytidine triphosphate (CTP). The main enzyme of this process is CTP synthetase. The chemical transformation involves the amination of the pyrimidine ring of the UTP molecule.

For this reaction to proceed successfully, two key components are required in addition to the substrate (uridine triphosphate) itself:

Overall equation for CTP synthesis by CTP synthetase: UTP + ATP + Glutamine → CTP + ADP + $P_i$ + Glutamate

General Scheme of UMP to CTP Conversion

The entire pathway from uridine monophosphate to cytidine triphosphate can be represented as a single biochemical reaction chain:

UMP → UDP → UTP → CTP

When analyzing this chain, it is important to remember the key components of each stage:

Mnemonic

To avoid confusing the enzymes, pay attention to the substrate: NMP kinase works with monophosphates (M), NDP kinase with diphosphates (D). The presence of the word "synthetase" in CTP synthetase always indicates a reaction that requires ATP energy.

Frequently asked questions

Which metabolic pathway provides the initial UMP for this synthesis?

The initial UMP comes from the de novo pyrimidine nucleotide biosynthesis pathway. This pathway includes six sequential steps and occurs predominantly in the cytosol. In these reactions, the pyrimidine ring is formed as orotic acid from glutamine, carbon dioxide, ATP, and aspartate. Then, orotate reacts with phosphoribosyl pyrophosphate to form orotidine-5'-monophosphate, the decarboxylation of which yields uridine monophosphate (UMP).

Which enzyme mediates the formation of deoxynucleotides from uridine and cytidine derivatives?

The formation of deoxynucleotides from uridine and cytidine derivatives is mediated by a key enzyme: ribonucleotide reductase (the ribonucleotide reductase complex). The process takes place at the diphosphate level (UDP and CDP). The mechanism involves the reduction of the ribose 2'-OH group to a hydrogen atom. Thioredoxin acts as the direct protein cofactor hydrogen donor for the reduction of ribonucleotides.

Which ATP residue is transferred during the formation of UDP and UTP?

During the phosphorylation of UMP and UDP, the $\gamma$-phosphate residue of the ATP molecule is transferred.

Which amino acid is the amino group donor during CTP synthesis?

Glutamine (Gln) acts as the donor, providing its amide group to aminate the pyrimidine ring.

Which enzyme catalyzes the conversion of UDP to UTP?

This reaction is catalyzed by NDP kinase (nucleoside diphosphate kinase).

What acts as the energy donor for the amination of the pyrimidine ring during CTP synthesis?

An ATP molecule serves as the energy donor for CTP synthetase.
